Knowledge of DNA sequences of genes and other parts of the genome of organisms has become indispensable for basic research studying biological processes, as well as in applied fields such as diagnostic or forensic research. The advent of DNA sequencing has significantly accelerated biological research and discovery. The rapid speed of sequencing attained with modern DNA sequencing technology has been instrumental in the sequencing of the human genome, in the Human Genome Project.

paa have been involved with DNA laboratories for over 15 years. paa were one of the original suppliers to the National DNA Database at the Forensic Science Service (FSS, UK).
The FSS were looking for piece of software that could control their small and large automated workcells, and OVERLORD Workstation (now Overlord3) was the chosen product. paa helped design and build the multiple automated workstations, both sample preparation and quantitation prior to amplification (left) and pre-analysis sample preparation prior to analysis by slab gel electrophoresis. Overlord is still used in the FSS laboratories for user instrument interaction, providing a standard software platform for all interactions with equipment in the laboratories. Overlord also provides complete sample and plate tracking to a central LIMS for legal and logistic purposes. |

Small-insert random DNA fragments are necessary to create efficient libraries for genomic sequencing projects. The success and efficiency of sequencing a large genome is dependent on the randomness of the fragments generated by the shearing of target DNA. Physical shearing methods (i.e., sonication, nebulization, and hydrodynamic shearing are preferentially chosen over enzymatic digestion due to the randomness and size of the fragments produced resulting in a suitable overlapping collection of fragments for sub-cloning.
